I wouldn’t start at 50. Try 5 rucks @20lbs 5@25 5@30 Etc. This will let you get your kit dialed in and your body used to the frequency. The Osprey will work. Just like backpacking the advice is to get the weight high and close to your back. This will take some fiddling with a large pack like yours.

I would study the suspension designs of the following backpacks for the shoulder strap, frame sheet part: Hill People Gear Umlindi, GoRuck GR1, GR2 or Rucker 4.0, Kifaru Checkpoint. Then I would look at the belt suspension systems of Kifaru larger packs, ULA Equipment and the belt Option for GoRuck.
